摘要
目的 :应用膜片钳技术研究了氯胺酮和羟丁酸钠联合应用时对钙激活钾通道的影响。方法 :实验选用培养 3~ 7天的神经元 ,在对称性高钾溶液中应用细胞吸附式和内面向外式构型记录通道电流。结果发现 :( 1)该通道电导为 170pS左右 ,对胞内Ca+敏感 ,特异性钾通道阻断剂能抑制或阻断该通道活动 ;( 2 )在细胞贴附式膜片上 ,氯胺酮和羟丁酸钠均明显增加该通道的开放概率 (Po) ,两药联合应用时的Po不是单独应用的简单相加 ,而具有较强的协同作用。结论 :氯胺酮和羟丁酸钠联合应用时对钙激活钾通道具有协同激活作用。
The purpose of this research was to study the effect of ketamine and sodium Gamma-hydroxybutyrate on the large conductance Ca 2+ -activated potassium (BK) channels in cultured hippocampal neurons of newborn rat.All the experiments were carried out on 3~7 days old neurons.Single channel current signals were recorded in symmetrical high K + solution (140mM) un der cell-attached patch and inside -out patch.The results showed that:(1)The unitary conductance was about 170pS,the BK channel activity was activated by internal calcium but was blocked by potassium channel blocker.(2)In cell-attached patch,when ketamine and sodium Gamma-hydroxybutyrate were added into the bath solution respectively,the open probability (Po) was markedly increased,however when ketamine was used in combination with sodium Gamma-hydroxybutyrate,the Powas not a simple addition of the valuse when they were used singalarly,but a result of coordination with each other.So,ketamine and sodium Gamma-hydroxybutyrate had a coordinated effect of Ca 2+ -activated potassium channels.
