羟丁酸钠对氯胺酮LD50和催眠ED50的影响

摘要 目的探讨羟丁酸钠(sodium oxybate,SO)对氯胺酮(ketamine,Ket)半数致死量(median lethal dose,LD50)和催眠半数有效量(median effective dose,ED50)的影响。方法生理盐水组(NS组)腹腔注射生理盐水10ml·kg-1,羟丁酸钠组(SO组)腹腔注射羟丁酸钠200mg·kg,20min后再分别腹腔注射等比剂量的氯胺酮溶液,用序贯法测定氯胺酮的LD50和催眠ED50。结果羟丁酸钠使氯胺酮LD50增高(P〈0.01),催眠ED50降低(P〈0.01)。结论羟丁酸钠能够降低氯胺酮的毒性,并可增强其催眠作用。 Objective To investigate the effects of sodium oxybate on the median lethal dose ( LDso ) and hypnotic median effective dose (ED50) of ketamine. Methods Intraperitoneal injection of normal saline ( 10 ml·kg -1 ) was ad- ministrated in normal saline (NS) group and sodium oxybate (200 mg· kg -1) was intraperitoneally injected in sodium oxybate (SO) group, followed by peritoneal injection of katamine in geometric proportion 20 min later. LDs0 and hypnotic EDso were measured by sequential method. Results Sodium oxybate promoted LD50 of ketamine ( P 〈 0.01 ) and de- pressed hypnotic ED50 ( P 〈 0.01 ). Conclusion Sodium oxybate can reduce the toxicity and reinforce the hypnotic effect of ketamine.
