补脾益气方对脾虚哮喘大鼠肺组织黏蛋白5AC表达的影响 被引量:11

摘要 目的观察补脾益气方治疗前、后的脾虚哮喘大鼠黏蛋白(mucin,MUC)5AC表达的变化,旨在探讨补脾益气方对脾虚哮喘气道黏液高分泌的影响。方法 50只W istar雄性大鼠,按照随机数字法分为5组:对照组、脾虚哮喘组、哮喘组、脾虚哮喘治疗组和哮喘治疗组,每组10只。采用ELISA测定支气管肺泡灌洗液(BALF)中MUC5AC含量,采用RT-PCR测定肺组织MUC5AC的mRNA表达水平。结果脾虚哮喘组和哮喘组大鼠BALF中MUC5AC含量和肺组织MUC5AC表达均显著升高,和对照组比较差异显著(P<0.01)。并且和哮喘组相比较,脾虚哮喘组BALF中MUC5AC含量和肺组织MUC5AC表达显著升高(P<0.01)。和脾虚哮喘组比较,脾虚哮喘治疗组BALF中MUC5AC含量和肺组织MUC5AC表达显著降低(P<0.01),和哮喘组比较,哮喘治疗组BALF中MUC5AC含量显著降低(P<0.01)。结论脾虚哮喘大鼠肺组织MUC5AC表达进一步升高,BALF中的MUC5AC含量进一步增加,补脾益气方能够使其不同程度逆转。 Objective To investigate the effect and mechanism of invigorating the spleen to benefit qi prescriptions on airway mucus hypersecretion.Methods 50 male Wistar rats were divided into 5 groups randomly: control,spleen-deficiency asthmatic model,asthmatic model,spleen-deficiency asthma treatment and asthma treatment groups.MUC5AC content were detected in bronchia alveolus lavage fluid(BALF) were measured by ELISA.MUC5AC mRNA expression level was detected by RT-PCR.Results The result from MUC5AC content and MUC5AC gene expressions in rats showed that compared with the control group,the two items were increased obviously in both spleen-deficiency asthma and asthma group(P0.01).The MUC5AC gene expression level in spleen-deficiency group was higher than that of asthma group(P0.01).After treatment,gene expression in these two groups were lower than before(P0.01).Conclusions The spleen deficiency may increase MUC5AC content in BALF and MUC5AC expression of lung in asthmatic rat models,but invigorating the spleen to benefit qi could reverse the above-mentioned items.
