摘要
目的:建立HPLC法测定藏药二十五味珊瑚丸中乌头碱的限量。方法:采用Phenomenex Gemini DOS(4.6mm×250mm,5μm)色谱柱,以0.04mol·L^-1三乙胺(用磷酸调节至pH3.0±0.1)-甲醇(60:40)为流动相,流速1.0mL·min^-1,检测波长235nm,柱温40℃。结果:乌头碱在1~120μg范围内,存在良好的线性关系(r=0.9998);检测限为10ng。结论:本方法快速简便,准确可靠,可用于二十五味珊瑚丸的质量控制。
Objective: To establish the HPLC method of determing aconitine in Ershiwuwei Shanhu Pills. Methods: The Phenomenex Gemini DOS ( 4. 6 mm×250mm,5μm)was used with a mobile phase of triethylamine(adjusted to pH 3.0±0. 1 by phosphoric acid )-methanol(60: 40), the wavelength of detection was set at 235 nm, the flow rate was 1.0 mL ·min-1 with column temperature at 40℃. Results: The calibration curves were linear in the rang of 1 - 120 μg ( r =0. 999 8) for aconitine. The limits for detection were 10 ng. Conclusion: The method is simple, reliable and reproducible and can be used effectively for the quality control of Ershiwuwei Shanhu Pills
