摘要
目的 研究大鼠肝星状细胞( H S C) L D L、 H D L 受体。方法 用链霉蛋白酶和胶原酶原位灌流, Nycodenz 密度梯度离心分离大鼠 H S C;应用125 I L D L 和125 I H D L3 配体进行放射性配基结合实验测定 H S C L D L、 H D L 受体。结果 H S C 膜表面存在 L D L、 H D L 受体, H S C 与浓度递增的125 I L D L 和125 I H D L3 共同孵育进行饱和试验, H S C L D L、 H D L 受体特异性结合 L D L、 H D L 的浓度效应曲线,呈可饱和性指数曲线,将特异结合用于 Scatchard 作图,r 分别为- 0 .9751 和- 0 .9616 ,解离常数分别为8 .4736n mol/ L 和9 .1256n mol/ L,最大结合容量分别为439 .32 和259 .34fmol/mg 蛋白。结论 H S C L D L、 H D L 受体对脂蛋白代谢及胆固醇的调节有重要作用,为阐明脂肪肝肝纤维化的发生提供了新的实验依据。
Objective To study the low density lipoprotein(LDL) and high density lipoprotein(HDL) receptors in hepatic stellate cell (HSC) from rat liver. Methods HSC were isolated and cultured from liver of Wistar rats by in situ perfusion with pronase and collagenase and density gradient centrifugation with Nycodenz. Radioligand conjugation assay with 125 I LDL and 125 I HDL 3 were detected for LDL and HDL receptors of HSC.Results It was demonstrated that there existed LDL and HDL conjugation sites on the rat HSC. On the basis of LDL and HDL receptor saturation analysis, Scatchard curve was obtained. K d and B max of 125 I LDL and 125 I HDL 3 bound to HSC were 8.4736 and 9.125nM, and 439.32 and 259.34fmol/mg cell protein, respectively.Conclusion LDL and HDL receptors of HSC might have an important role in the metabolism of lipoprotein and regulation of cholesterol. These results provided the basis of important theory and experimentation for the relationship between fatty liver and liver fibrosis.(Shanghai Med J, 1999,22∶589 591)
