硒抑制人癌细胞株和人B细胞的EB病毒效应

摘要 应用间接免疫荧光法和显微荧光分光光度技术,研究了试管内无细胞毒性浓度的亚硒酸纳(硒)对来源于伯基特淋巴瘤的Raji细胞株,EpsteinBar病毒(EBV)抗原表达的影响以及抑制EBV转化人脐血B淋巴细胞(BL)的作用。结果表明,001~01mg/L的硒使Raji细胞的EA抗原抑制353%~501%;001~05mg/L的硒令EBNAMFS值降低84%~218%;01~10%mg/L的硒明显抑制EBV转化BL,抑制率为633%~921%。由于EBV与鼻咽癌(NPC)密切相关,NPC病人血清和头发中硒含量明显低于健康人。本研究提示:对NPC患者和EBV基因表达活跃的人群适当补硒,可能有助于NPC和与EBV相关疾病的预防和辅助治疗。 The effects of non toxic doses of sodium selenite(Se)on the expression of Epstcin Barr virus (EBV) antigens in Raji cell line originated from Burkitt's lymphoma were studied by indirect immunofluorescence staining and micro fluorescence spectrophoto quantitation (MFS),as well as the inhibiting transformation in B lymphocytes from human umbilical cord blood (BL) with EBV in vitro by Se. It was found that the early antigen (EA)was inhibited 35.3%~50 1% by Se (0.01~0.1 mg/L) and Se (0.01~0.5mg/L)can lower nnclear antigen (EBNA) MFS value for 8.4%~21.8% in Raji cell linc.The transformation effect of BL with EBV was inhibited by Se (0.1~1.0mg/L).The inhibition rate of BL transformation was 60.3%~92.1%.EBV is associated with the nasopharyngeal carcinoma(NPC).The Se levels in serum and hair of patients with NPC were lower than that of healthy persons.The results suggest that supplement Se to patients with NPC and persons for active expression of EBV gene may protect against and treat NPC and diseases associated with EBV.
