摘要
通过基因重组,构建携带MLV启动子及EB病毒瘤基因BNLF-1的融合基因,经显微注射后共产生原代转基因小鼠12只,这些小鼠在生长发育上与同胞胎中的野生型小鼠并无显著差异,但其发育期死亡率却高得多(9/12).对死亡小鼠的组织病理学诊断表明:肺、脾、肝、肾等器官均有程度不同的病变,有5只小鼠可确定为假结核病,存活的3只中有1只眼部感来致盲。上述结果提示,这种小鼠可能因免疫系统异常而易受致病微生物感染。
fusion gene carrying promoter/inhancer sequence of MLV and BNLF-1 gene of EB viruswas constructed with the method of gene recombination. Twelve positive transgenic mice weregenerated by the fertilized egg microinjection technique. There was no evident difference in developing between the positive transgenic mice and wild-type siblings,but the positive founder micehad a higher death percentage. Histopathologic examination showed:there were pathologicalchange in the. tissues of lung,liver,spleen and kidney.Five founder transgenic mice died ofpseudotuberculosis. The results indicate that the BNLF-1 transgenic mice may be a dysimmuni-ty,which results in poor resistance to infection of pathogenic bacteria.
