摘要
阿魏酸在溶剂中不稳定,这给定量分析带来一定干扰.研究了阿魏酸在6种不同溶剂系统中的稳定性,改进了溶解阿魏酸的溶剂,在此基础上建立了生药当归中阿魏酸的高效毛细管区带电泳分离分析方法.电泳条件:以60mmol/L硼砂(pH9.45)为电泳介质,未涂层弹性融硅毛细管(50μm×39.5cm,有效分离长度34.8cm)为分离通道,压力进样(68.95kPa·s),20kV恒压电泳(25℃),检测波长314um.实验发现,在4mg/L~400mg/L范围内,阿魏酸可进行定量分析,加样回收率为100.4±1.2%.方法简便快速、结果准确,重现性好,可用于当归及其制剂的质量控制.
Ferulic acid is unstable in solution and it is easily decomposed under the light. In this article stability of ferulic acid in six different kinds of solutions was investigated with UV spectrophotometry, based on the results, a method was established to separate and determine ferulic acid in Redix Angelicae Sinensis(Danggui) by HPCE. Electrophoretic conditions:uncoated fused silica capillary (34.8/39.5 cm,50μm i.d) , 60 mmol/L borax(pH 9.45) , appb'ed voltage 20 kV, detection 314 nm, injection 68.95kPa·s. Under the optimum conditions, ferulic acid was separated successfully from other componentswithin 15 minutes. The relative peak area of ferulic acid increases linearly with the increase of itsconcentration in the range of 4 mg/L~400 mg/L, and the recovery is 100.4%±1.2%. The analyticalresults demonstrated the method was simple, rapid and well reproducible, and could be used as a reliable tool for the quantity control of danggui and its descriptions.
