C肽光激化学发光免疫分析试剂盒的研制 被引量:5

摘要 目的:研制光激化学发光免疫分析技术(AlphaLISA)建立人血清C肽(C peptide)的快速检测试剂盒。方法:采用夹心法建立C peptide AlphaLISA试剂盒。结果:自制C肽试剂盒灵敏度为0.06ng/ml.,可测范围为(0.06~22)ng/ml。批内与批间的精密度分别为4.0%~4.8%,5.6%~6.8%,与胰岛素以及胰岛素原无交叉反应,使用本试剂盒与CMIA-ARCHITECT试剂盒同时检测98份临床血清样本,结果具有相关性,其相关系数为0.973。结论:自制C肽AlphaLISA试剂盒的各项性能指标能够达到临床检测要求,可用于临床血清样本C肽浓度的测定。 Objective To establish the amplified luminescent proximity homogeneous immunoassay(AlphaLISA) kit fordetection of human C peptide.Methods The double antibody sandwich method was used to develop the AlphaLISA kit for detection of serum C peptide.Results The working range of the kit was 0.06~22ng/ml.The sensitivity was reached to 0.06ng/ml.The intra-and inter-assay coefficient of variation were 4.0%~4.8% and 5.6%~6.8%,respectively,moreover.There was no cross-reaction with insulin or proinsulin respectively.98 samples were tested by the two kinds of self-made and commercially available CMIA(ARCHITECT) kits at the same time.The correlation coefficient was 0.973.Conclusion The self-made AlphaLISA kit for C peptide detection could meet the clinical requirements for the detection of the concentration.
