高温对黑色素瘤细胞的影响——黑色素生成及DOPA反应组化观察

摘要 本文采用显微荧光分光仪测定法及3,4二羟基苯丙氨酸(DOPA)反应的超微组化方法,观察高温对S91黑色素瘤细胞的影响。结果:与对照组比较,细胞经42℃(3小时)或44℃(1小时)加温后,黑色素含量显著减少(P<0.01)。经0.1%DOPA孵育电镜下显示:42℃加温组,胞浆内各型黑色素小体减少,Ⅰ、Ⅱ型尤为明显。DOPA反应阳性产物消失。高尔基氏器、线粒体及细胞核亦受到损害;44℃加温组,细胞内出现大量空泡,结构模糊,不易辨认。 The effects of hyperthermia on melanin content and DOPA-reaction of S91 melanoma cells in vitro were studied by means of fluorometric assay and ultra-histochemical method.The results suggested that hypeithermia (42℃ for 3h or 44℃ for 1h) may cause melanin content significantly reduced compared with that of cells growing under normal condition (37℃) (P<0.01).The cells treated with 42℃ showed that different types of the melanosomes,especially types 1 and 2,were markedly decreased.No DOPA-positive reaction was noted.Mitochondria,Golgi organe-lles and nucleus were damaged in varying degrees.At 44℃ the cells became blurred and there were numerous vacuoles in cytoplasm
