摘要
以高抗品种东农9674 为父本, 以高感品种东农87-104 为母本, 杂交得到F2 代群体。该群体经人工接种灰斑病1号生理小种后, 分别从抗、感材料中取15 株的叶片提取DNA组成抗、感池, 用820 个10m er随机引物进行RAPD多态性分析, 其中3个引物在抗、感池间出现稳定的多态性标记OPK03840, OPM171700, OPO10950, 并在F2 代个体中表现出抗性与多态性标记协同分离的趋势, 这三个标记与抗性基因Rf1 的连锁顺序为OPK03840-Rf1-OPM171700-OPO10950, 连锁距离分别为10.4cM-Rf1-13.8cM-26.1cM。
A F 2 population was generated from a cross between DongNong 9674(the resistant parent) and DongNong 87 104 (the susceptible parent). Following the approach of bulked segregant analysis, eight hundred and twenty RAPD primers were screened to identify the markers linked to the reststance gene Rf1 of Cercospora Sojina race1 in soybean. Three RAPD markers OPK03 840 , OPM17 1700 and OPO10 950 , linked to the resistance gene, were selected and mapped by analyzing F 2 population with the software MAPMARKER II. The positions of the markers relative to the target gene Rf1 are OPK03 840 Rf1 OPM17 1700 OPO10 950 , and the distances are 10 4cM Rf1 13 8cM 26 1cM.
