摘要
目的:探讨具有雌激素活性的中药单体异补骨脂素(ISR)对过氧化氢(H2O2)诱导氧化损伤的人晶状体上皮细胞(HLEC)NF-κB表达的影响。方法:实验分正常组、H2O2组、雌二醇(E2)组、异补骨脂素(ISR)组、核因子-κB(NF-κB)抑制剂二硫代氨基甲酸吡咯烷(PDTC)组,流式细胞术(FCM)测定上述5组标本中HLEC的NF-κB的表达率。结果:以上各组,NF-κB的平均荧光表达率分别为:9.53%、39.87%、25.18%、23.23%、5.90%;正常HLEC能表达一定的NF-κB,H2O2组NF-κB的表达率明显增高,NF-κB抑制剂PDTC组的NF-κB的表达率明显降低,ISR组和E2组的NF-κB的表达率则介于以上两组之间。结论:H2O2可以诱导人晶状体上皮细胞NF-κB的活化表达,具有雌激素活性的中药单体ISR可以有效抑制NF-κB的活化表达,其抗HLEC氧化损伤作用可能是通过下调NF-κB表达实现的。
Objective:To investigate the influence of isopsoralen(ISR) with estrogenic activity on expression of nuclear factor-kappa B(NF-κB) in HLEC afer oxidative damage induced by H2O2.Methods:The cultured HLEC were divided into 5 groups:control group,H2O2 group,β-Estradiol(E2) group,ISR group and PDTC(NF-κB inhibitor) group.The expression of NF-κB in HLEC was detected by flow cytometer(FCM).Results:In the above groups,the mean fluorescence expression rate of NF-κB were:9.53%,39.87%,25.18%,23.23%,5.90%.The normal HLEC express NF-κB;The expression rate of NF-κB in H2O2 group was increased obviously compared with control group;The expression rate of NF-κB in PDTC(NF-κB inhibitor) group was decreased conspicuously compared with H2O2 group;The expression rates of NF-κB in ISR and E2 group were between H2O2 group and PDTC group.Conclusion:The activation of NF-κB in human lens epithelial cells could be induced by H2O2.ISR with estrogenic activity could inhibit the activation of NF-κB,the down-regulated NF-κB may be involved in the role of Antioxidant damage.
