摘要
目的探讨肿瘤抑制基因ｐ５３、ＭＧＭＴ在急性非淋巴细胞白血病（ＡＮＬＬ）中的改变。方法用聚合酶链反应单链构象多态性（ＰＣＲＳＳＣＰ）技术对３１例ＡＮＬＬ患者、２２例正常人ｐ５３基因第５、６、７、８外显子和ＭＧＭＴ基因第１、４外显子进行突变分析。结果３１例ＡＮＬＬ中５例存在ｐ５３基因突变，４例存在ＭＧＭＴ基因突变，其中２例ｐ５３与ＭＧＭＴ同时发生突变。结论ｐ５３基因突变在白血病发生中起一定作用。ＭＧＭＴ的作用有待进一步研究。
ObjectiveTo investigate the incidence of p 53 and MGMT gene alteration in acute non lymphocytic leukemia(ANLL).MethodsUsing polymerase chain reaction single stand conformation polymorphism(PCR-SSCP) technique,31 cases of acute non lymphocytic leukemia(ANLL) and 22 normal controls were studied for mutation in p53 gene exons 5,6,7,8 and MGMT gene exons1,4.ResultsIn 31 cases of ANLL,4 cases showed MGMT gene mutation,5 cases p53 gene mutation and 2 cases both p53 and MGMT mutation.Conclusionp53 gene mutation may play an important role in the pathogenesis of acute leukemia,while the role of MGMT gene mutation remains to be elucidated.
