摘要
采用聚合酶链反应-单链构象多态性(PCR-SSCP)法对41例脑肿瘤进行P53基因突变的检测,并与抗突变型P53蛋白单抗的免疫组化染色结果相对照,结果显示脑肿瘤中P53基因突变率为34.1%(14/41),胶质瘤为28%(10/36)。P53基因突变与突变型P53蛋白表达具有高度一致性,且二者均与脑肿瘤的分化程度有关。P53基因突变及蛋白的表达不仅出现在高恶度胶质瘤及转移癌,而且可出现在低恶度胶质瘤中,表明P53基因的突变在脑瘤的发生、发展过程中起重要作用。
P53 gene mutation in 41 brain tumors were analyzed by single-strand conformation polymorphism(SSCP),and the results were compared with P53 protein expression detected by immunohistochemistry.P53 gene mutations were found in 14 out of 41 brain tumors(34.1%),in 10 of 36 gliomas(28%).There was a highly significant association between the presence of P53 mutations and P53 nuclear reactivity(P<0.001).P53 alteration(mutation and expression)were related to grading of malignancy of brain tumors.P53 gene mutation was found not only in high-malignant glioma,but also in low-malignant glioma.This indicates that P53 gene plays an important role in the development transformation of brain tumors,and malignant tumors.
