摘要
目的:了解接种患者血清的恒河猴能否复制 T T 病毒( T T virus, T T V),正常恒河猴中是否有 T T V 感染,分析猴中 T T V 基因结构特点。方法:用 P C R 方法检测血清标本中的 T T V D N A,对 T T V D N A 阳性的标本进行序列测定。结果:健康猴中无 T T V 感染;10 只接种患者血清的恒河猴中,经检测有 4 只为 T T V 阳性。将其中一株 T T V 进行序列测定,该序列与日本 T T V 部分基因相对应位置的核苷酸同源性为 96% ,与中国株 T T V 全基因相对应位置的核苷酸同源性为 98% 。结论:健康猴群中无 T T V 自然感染。恒河猴可感染 T T V,可作为研究 T T V 的动物模型。
Objective: To learn whether natural infection of TTV exists in Macaca mulatta or not, and to investigate the replication of TTV in Macaca mulatta inoculated with serum of patients with non A E hepatitis. Methods: Serum samples collected from inoculated and normal monkeys were examined for TTV DNA using PCR method and the PCR products were sequenced. Results and Conclusion: TTV DNA was positive in 4 of 10 inoculated monkeys and none in normal monkeys. The partial gene of TTV was cloned and sequenced. The partial gene of TTV isolated from monkey showed 96% nucleotide identity with the corresponding one of TTV isolated in Japan, and 98% identity with TTVCH1(isolated in China).
