肝细胞去唾液酸糖蛋白受体介导的超顺磁性氧化铁粒子磁共振增强研究 被引量:3

摘要 目的:探讨标记乳糖基白蛋白超顺磁性氧化铁粒子(LAC-HAS-SPIO)与肝细胞去唾液酸糖蛋白受体(ASG)的结合特性。方法:采用NaBH3cN还原法制备富含乳糖基的白蛋白(LAC-HSA),然后将其标记上粒径18.6±19um的SPIO(LAC-HAS-SPIO)通过体外及体内试验,观察ASG受体阻断剂D一半乳糖对LAG-SPIO增强效果的影响;电镜观察和比较LAC-HAS-SPIO与普遍SPIO在肝内分布差异。结果:①电镜发现普通SPIO仅分布在kupffer细胞,而LAC-HAS-SPIO能分布到肝细胞内及肝细胞膜上;②预先加入D一半乳糖的试管,其信号强度远低于单纯加LAC-HAS-SPIO管;③大鼠预先注射D-半乳糖组,肝组织增强度为-60.3%±21.2%,低于来注射D-半乳糖组(肝强化率-89.4%±25.5%,0.01<P<0.05).结论:LAC-HAS_SPIO是一种新型的针对ASG受体的肝特异显像剂,对发现肝脏疾病的早期形态及功能变化将有重要价值。 Objective: To investigate the feasibility of 1actosaminaed human serum alhumin coated Superparamagnetie iron oxide Particles (LAGHAS-SPIO) binded specifically to hepaic asialoglycoproein receptor (ASG-receptor). Methods: LAG-HSA was Prepares by reductive drination method in the presence of NaBH3CN. Then, LAC-HSA was reacted with eding of Fe2+ and Fe3+ to synthsize.IAC-HAS-SPO. The contrast enhancement experiment of IAC-HASSPO were cacarried 0ut in vitro (in saxnples 0f the hepatic membrane suspensions enriched With ASG receptors) and in vivo (in 8 rats) with and withollt D-galactose (a competihve blocker to sabee ASG binding sites), respectively. The biwhstribution differenees between llsual SPIO and IAC-HAS-SPIO in rat livers were observed under electron ndcroscope. Results: ① Under electron thecroscope, electron dense Particles were identified only in kupffer cells in the usual SPIO group. on the contmp, large amouns of electron dease deposit were identified on the surface of hepaocyte mernbranes and within hepatic cytoplasms.②The signal intensity of hepeic membrane suspension was significaily lower after incubaion with LAC-HAS-SPIO after blockade using D-galactose. @The liver enhancing rate in the rats (n = 4) received Dgalactose hefore adrinishation 0f LAGHAS-SPIO,was - 6o.3% t 2l .2%,those reached - 89.4% l 25.5% in the rats (n = 4) which did not received injecton Of Dgalaclose.(0 .01 < P < 0. 05). Conclusions: The results indicate that LAC-HAS-SPIO is a hepatic Ase receptor directed contran agent. It may be clinicallly useful for monitoring early morphologic and functional changes in hepaic diseases.
