摘要
目的:建立C57BL/6小鼠脉络膜新生血管(Choroidal neovascularization,CNV)动物模型的评估方法。方法:①应用氪红激光对小鼠实验性视网膜进行光凝,激光的功率、光凝斑直径和曝光时间分别为250 mW、50μm及0.05 s诱导CNV动物模型。②分别于光凝后1、3、5、7、10d,用2×106FITC-dextran荧光素灌注小鼠心脏。③mAb特定抗Elastin抗体标记Bruch膜。④视网膜色素上皮(Retinal pigmentepithelium,RPE)-脉络膜-巩膜复合体铺片。⑤激光共聚焦显微镜对RPE-脉络膜-巩膜复合体进行CNV形态进行观察和测量。结果:光凝后3 d可见新生血管及走向清晰可见,光凝后7 d CNV达到高峰。结论:用抗体标记Bruch膜。FITC-dextran荧光素灌注小鼠心脏,激光共聚焦显微镜对RPE-脉络膜-巩膜复合体进行氪红激光诱导C57BL/6小鼠的CNV模型进行评估,直观,可靠。
Objective: To establish a new assessment method for animal model of choroidal neovascularization(CNV).Methods: In this model,CNV was induced by laser photocoagulation in C57BL/6 mice with the krypton red laser.Animals from each group were sacrificed and were perfused through the heart with fluorescein-labeled dextran(FITC-Dextran) on day 1,3,5,and 7 post laser.Retinal pigment epithelium-choroid-scleral tissue,after using a mAb specific for elastin staining,was examined to determine the incidence and size of CNV complex by confocal microscopy.Results: Confocal micrograph of the neovascular complex in C57BL/6 mice showed the new vessels(green),and damaged Bruch's membrane(BM) and RPE were stained red for elastin.CNV appeared on day 3 post laser,reaching the peak on day 7 post laser.Conclusion: The new assessment method of animal model of choroidal neovasculation(CNV) is feasible,reliable and visualized.
