应用视网膜铺片和神经元逆行标记技术测定神经节细胞密度分布 被引量:6

Determination of density distribution in retina ganglion cell using methods of retinal preparation and retrograde cell labeling of carbocyanine dye

摘要 目的:为视网膜神经节细胞(retina ganglion cell,RGC)的准确定量研究提供形态学的指标依据。方法:联合应用Nissl染色法和神经元逆行标记技术,并通过计算机图像处理技术测定RGC的形态分布特征。结果:两种标记方法在视网膜组织中标记的细胞形态、大小和密度分布呈现明显的差异。Nissl染色可以使所有细胞着色,神经元逆行标记技术仅使RGC着色。通过两者的综合分析,在RGC等密度曲线图中可以观察到在视神经乳头下方形成一个沿鼻颞侧轴方向伸展的高密度区,即视条纹,由视条纹至周边部细胞密度递减。结论:联合应用视网膜铺片法和神经元逆行标记技术两者的优点,能够较准确地测定RGC的密度、大小及其分布等形态特征。 AIM: To provide the functional status for the quantitative studies of retinal ganglion cell (RGC). METHODS: The combined methods of Nissl staining, retrograde labeling of carbocyanine dye and image analysis by a computer were used. RESULTS: The total cells in the ganglion cell layer were stained in Nissl retinal preparation, while only RGC were labeled in DiI-labeled preparation. From the analysis of Nissl staining and retrograde labeling of carbocyanine dye, a horizontal region of high-density (visual streak) was found in the central retina below the optic disk. The cell density decreased from the visual streak to the peripheral zone. CONCLUSION: The combined methods can be characterized the size and density distribution of ganglion cells in the ganglion cell layer.
