摘要
目的:对横根费菜的组织形态进行鉴别,并建立横根费菜中没食子酸含量测定方法。方法:采集原植物,对该植物进行植物学及生药学的鉴定;采用HPLC-UV法,色谱柱为Hypersil C18柱(4.6 mm×250 mm,5μm);流动相:甲醇-体积分数为0.3%磷酸水溶液(94∶6);检测波长:273 nm;流速:1.0 mL/min。结果:横根费菜的生药学特征明显。根状茎木质,横生有分枝;叶横切面上、下表皮细胞气孔多为不等式、环式,叶脉维管束为周木型;茎维管束外韧型,茎的表皮层和髓较宽阔,导管多为螺纹,可见网纹。没食子酸在0.12~1.20μg质量范围内呈良好线性关系(r=0.999 9)。结论:横根费菜生药学特征明显,易于观察;没食子酸的含量测定方法简单、快捷、准确,为制定横根费菜药材质量标准提供依据。
Objective:To identify the tissue structure of Sedum kamtschaticum Fish,and establish a method for the determination of gallic acid in Sedum kamtschaticum Fish.Methods:The plant was collected to study on the botanical and pharmacognostical characteristics.The determination by HPLC was performed on a hypersil column C18(4.6 mm×250 mm,5 μm) by UV detector at 273 nm with methanol-0.3% phosphoric acid(94:6) as the mobile phase,and the flow rate was 1.0 mL/min.Results: Sedum kamtschaticum Fish was of pharmacognostical characteristics of rhizome lignin,diageotropism with branches;the inequality and concentric type in most of leaf cross section top and bottom epidermal cell blowhole;amphivasal leaf vein bundle;collateral vascular bundle,broad stem's cuticular layer and the marrow,thread of plant duct with obvious cobwebbing.The calibration curve showed a good linearity within the range of 0.12~1.20 μg(r=0.999 9).Conclusion: The pharmacognostical characteristics of Sedum kamtschaticum Fish are obvious.The method is proved to be simple,accurate which provides the basis for controlling the quality of Sedum kamtschaticum Fish.
