中医不同治法对大鼠心肌缺血再灌注损伤后细胞凋亡的影响 被引量:12

摘要 目的观察中医不同治法补气、活血、化痰对大鼠心肌缺血再灌注(I/R)诱导细胞凋亡及凋亡相关基因Bc1-2、Bax和氧自由基含量的影响。方法建立大鼠动脉粥样硬化在体心肌缺血再灌注模型,采用原位末端TUNEL法检测心肌细胞凋亡指数(AI),链酶亲和素-生物素-酶复合物(SABC)免疫组化法检测凋亡相关基因Bcl-2、Bax蛋白表达,常规方法检测丙二醇(MDA)含量、超氧化物歧化酶(SOD)活性。结果与I/R组比较,中医补气、活血、化痰法均能降低AI,有效清除氧自由基,差异有统计学意义(P<0.01);与I/R组比较,其中活血法和化痰法均能上调Bc1-2表达、下调Bax表达,差异有统计学意义(P<0.01),最终达到保护I/R心肌的目的。结论中医不同治法补气、活血、化痰可能通过不同途径预防I/R心肌细胞发生凋亡,从而对I/R心肌具有保护作用。 Objective To observe the effect of different treatments in TCM on myocyte apoptosis and expression of Bc1-2 and Bax and the content of oxygen free radicals in myocardial ischemia reperfusion in rats.Method The rats models in vivo of atherosclerosis myocardial ischemia reperfusion were stablished,the apoptosis index(AI) was detected withTUNEL method in situ end method,the apoptosis related genes Bcl-2Chain and Bax protein expression with avidin-biotin-enzyme complex(SABC) immunohistochemical method,and the activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) with routine method.Results Compared with I/R group,all the treatments in TCM could reduce AI and remove effectively the oxygen free radicals(P0.01),the activating blood and resolving phlegm methods could reduce the content of MDA and increase the activity of SOD and decrease Bax expression(P0.01),and then,protect 1/R cardiac muscle.Conclusion The different treatments in TCM can restrain the apoptosis of I/R cardiac muscle cells and to protect consequently the I/R cardiac muscle.
