蝴蝶兰离体培养花梗表面消毒试验初报 被引量:2

A Preliminary Report on Surface Sterilization of Butterfly Orchid Pedicels in Vitro

摘要 以蝴蝶兰花梗为试验材料,进行花梗外植体表面消毒处理研究,初步建立了一套适合蝴蝶兰离体培养花梗表面消毒的体系。研究结果表明,(1)H2O2处理2 d后都出现污染,以细菌污染为主,7 d后污染率达100%;NaClO以40 min处理的效果较好,污染率为88.23%;升汞消毒以25min消毒效果最好,污染率仅为13.33%;(2)二次消毒法出现污染的时间为处理后6 d,污染率仅为13.33%,酒精浸泡法和一次性消毒法均在处理后4 d出现污染,污染率分别为83.33%和46.66%。上述结果表明,升汞的消毒效果优于NaClO和H2O2,二次消毒法优于酒精浸泡法和一次性消毒法,初步建立了蝴蝶兰花梗的消毒体系为升汞消毒2次。 Taking butterfly orchid pedicel as materials, sterilization system for butterfly orchid pedicel was investigated. The results showed that, (1) bacterial contamination appeared after 2 days in the H2O2 treatment, and the contamination rate reached 100% after 7 days. The optimal time of NaClO treatment was 40 rains, and the contamination rate was 88.23% after 2 days. The best time of mercuric chloride treatment was at 25 min, and the contamination rate was only 13.33% after 6 days. (2) The contamination rate was only 13.33% after 6 days with the secondary sterilization treatment, but the contamination rates of alcohol soaking and one-time sterilization were 83.33 % and 46.66 % after 4 days treatment respectively. Above mentioned results indicated that the sterizating effect of mercuric chloride was better than that of NaClO and H2O2, and the secondary sterilization was better than alcohol soaking and one-time sterilization. The preliminary sterilization system for butterfly orchid pedicel was established, which was to use mercuric chloride treatment for 25 mins at twice.
