摘要
目的:研究酪蛋白磷酸肽钙磷复合体(Casein phosphopeptide-Amorphic Calcium Phosphate,CPP-ACP)对发酵乳杆菌生长、粘附的影响,进一步探讨CPP-ACP的防龋机制。方法:将发酵乳杆菌接种到BHI培养基,实验组中加入不同浓度(0.5%~5.0%(W/V))的CPP-ACP溶液,厌氧培养48 h后,采用噻唑蓝(MTT)比色分析法测定细菌浓度的吸光度A值(λ=550nm)。采用唾液包被的羟磷灰石(Saliva-coated hydroxyapatite,S-HA)形成实验性膜,用不同浓度(0.5%~5.0%(W/V))CPP-ACP处理S-HA,定量观察发酵乳杆菌在S-HA上的粘附情况。结果:随CPP-ACP浓度的升高,发酵乳杆菌甲臜产物的二甲亚砜溶液的吸光度值降低,即发酵乳杆菌的活菌数减少(P<0.01)。发酵乳杆菌对经各实验浓度的CPP-ACP处理后的S-HA粘附能力明显下降,粘附量(cpm)随CPP-ACP浓度的升高而降低,粘附抑制率达100%。结论:CPP-ACP对发酵乳杆菌的生长以及在S-HA上的粘附具有抑制作用,随CPP-ACP浓度的升高抑制作用增强。
Objective:To investigate the effects of casein phosphopeptide-amorphic calcium phosphate(CPP-ACP) on the growth and adherence of Lactobacillus fermentum and the possible anti-caries mechanism of CPP-ACP.Methods:Lactobacillus fermentum was inoculated in BHI uith different concentrations of CPP-ACP and grew in anaerobic condition at 37℃ for 48 hours.MTT method was used to measure the effects of CPP-ACP on the growth of Lactobacillus fermentum.The hydroxyapatites(HA) beads mere coated with saliva to form saliva-coated hydroxyapatite(S-HA),as in vitro adhesion model of experimental pellicle.Lactobacillus fermentum was radiolabeled with 3H-thymidine and different concentrations(0.5%~5.0%(W/V))of CPP-ACP pretreated S-HA.The numbers of 3H-thymidine labeled bacteria attached to S-HA were counted by scintillation counting.Results:It was found that CPP-ACP could inhibit the growth and adherence of Lactobacillus fermentum.With the concentration of CPP-ACP increasing,the values of A(550nm) and cpm decreased(P〈0.01),and all of the inhibition rate of adherence arrived at 100%.Conclusion:CPP-ACP had inhibitory effect on the growth and adherence of Lactobacillus fermentum,the inhibitory effect enhanced with the increasing of CPP-ACP concentration.
