摘要 组合生物合成是通过对微生物代谢途径中一些酶的编码基因进行操作,从而获得许多新的"非天然"天然产物。本文重点介绍聚酮合酶(PKSs)和非核糖体多肽合酶(NRPSs)的分类、作用机制及其杂合系统在组合生物合成应用研究中相关报道。另外,对于PKSs和NRPSs在大肠埃希菌中的异源表达也作了简单介绍。 Combinatorial biosynthesis can produce numerous new "unnatural" natural products through manipulating genes encoding enzymes in microbial metabolic pathway. This article reviews the type, the mechanism of polyketide synthases (PKSs) and nonribosomal peptide synthases (NRPSs) as well as the application of acquiring new polyketides (PKs), nonribosomal peptides (NRPs) and novel compounds resulting from hybrid PKSs/NRPSs through combinatorial biosynthesis. Besides, the expression of PKSs and NRPSs in Escherichia coli will also be introduced.
