摘要
1,2,3-三取代四氢-β-咔啉化合物与胺在DMSO中反应制得他达那非及其类似物;不经处理,在1,8-二氮杂双环[5,4,0]十一碳-7-烯存在下完成C-12a-位差像异构化,高立体选择性地一锅合成了他达那非12 a-位差向异构体及其类似物,其结构经1H NMR,IR,MS和元素分析表征。
12a-epi-Tadalafil and its analogues were synthesized by the reaction of 1,2,3-trisubstituted tetrahydro-β-carboline with amines in DMSO,then C-12a-epimerization in the presence of DBU in one-pot.The structures were characterized by 1H NMR,IR,MS and elemental analysis.
