摘要
目的 研究结肠癌中p16 基因的失活方式.方法 采用Southern blot 法鉴定了结肠癌细胞株SW480 ,Colon205 ,HR8348 ,HT29 ,Colo320 ,LSIGT9695 及18 例正常结肠粘膜组织中p16 基因的甲基化情况.结果 在6 株结肠癌细胞株中有5 株(83-3 % ) p16 基因CpG岛呈异常甲基化状态,与之相对,18 例正常结肠粘膜组织中仅5 例(27-8 % ) 表现为异常甲基化.结论 在结肠癌中p16 基因因异常甲基化而失活,这为结肠癌的基因治疗提供了新策略。
AIM To study the inactivation mode of p16 gene in colon cancer. METHODS The methylation rate of p16 gene in colon cancer cell lines (Colo205 , HT29 , SW480 , HR8348 , Colo 320 , LSIGT9695) and 18 cases of normal colonic tissues were examined by using Southern blot method . RESULTS The aberrant methylation of 5○CpGisland of p16 gene was found to be positive in 5 of 6 (83-3 % ) colon cancer celllines.In contrast,only 5 of 18(27-8 % )normal colonic tissues showed aberrant methylation of the 5○CpG island of p16 gene . CONCLUSION The p16 gene in colon cancerisinactivated by aberrant methylation . This can provide a new tactics and a new targetfor gene therapy of colon cancer.
