摘要
以半番鸭血液基因组为模板进行PCR扩增,获得了半番鸭Agouti基因部分序列,该序列长为1178bp。序列分析表明该序列由部分第1外显子(92bp)、第1内含子(105bp)、完整的第2外显子(95bp)、完整的第2内含子(851bp)和部分第3外显子(35bp)组成;应用PCR-SSCP技术对扩增序列进一步研究发现位于第2外显子80bp处存在C/T的沉默突变,该突变的发现为进一步开展研究Agouti基因多态性与鸭羽色的相关性研究奠定基础。
A pair of primer was designed to amplify the sequence of Agouti gene from mule duck genomic DNA.The fragment was cloned and sequenced which was 1 178 bp in length.The sequence analysis showed this fragment consisted of partial exon 1(92 bp),intron 1(105 bp),exon 2 (95 bp),intron 2 (851 bp)and partial exon 3(35 bp).Using PCR-SSCP for this amplified fragment,a C/T silent mutant was found at 80 bp in exon 2,which is useful for the further study on polymorphism and the correlation on duck feather color.
