急性肺缺氧代谢对肺泡上皮Ⅱ型细胞超微结构、转化生长因子β1及凋亡诱导因子表达的影响 被引量:7

摘要 目的研究急性肺缺氧代谢对肺泡Ⅱ型上皮细胞(AEC-Ⅱ)超微结构、肺组织及血清中转化生长因子β1(TGF-β1)和凋亡诱导因子(AIF)表达的影响。方法 40只SD大鼠随机均分为A、B、C及D组,A、B、C组行单肺通气;D组行双肺通气。分别于实验30min(A组)、60min(B组)及120min(C和D组)测定肺组织超微结构变化以及血清和肺组织TGF-β1、AIF表达。结果 A、B及C组均出现了不同程度的AEC-Ⅱ细胞超微结构的改变,而D组基本正常。A、B、C组肺组织与血清中TGF-β1、AIF表达高于D组,且随缺氧时间延长呈上升趋势(D组<A组<B组<C组,P<0.01)。结论低氧代谢引起大鼠肺组织细胞结构发生改变及TGF-β1和AIF表达上调。 Objective To investigate the effects of acute pulmonary hypoxia metabolism on the ultrastructure of typeⅡalveolar epithelium cells(AEC-Ⅱ),transforming growth factor-β1(TGF-β1)and apoptosis inducing factor(AIF).Methods Forty SD rats were equally randomized into one of the four groups(n=10).Rats in group D underwent two lung ventilation while groups A,B,and C underwent single lung ventilation.Ultrastructure alteration of AEC-Ⅱand TGF-β1 and AIF levels in the serum and lung were evaluated at 30(group A),60(group B),and 120 min(groups C and D)after ventilation,respectively.Results Alterations of AEC-II were observed in groups A,B,and C but differed in various degrees while there was virtually no changes in group D.Lung tissue and serum levels of TGF-β1 and AIF were significantly higher in groups A,B and C than those in group D with the following sequence(D 〈A〈 B 〈C,P〈0.01).Conclusion Acute hypoxia induces alterations of AEC-Ⅱand higher levels of TGF-β1 and AIF expression.
