补肾中药对帕金森病模型小鼠黑质-纹状体神经元的保护作用 被引量:14

摘要 目的探讨补肾中药对帕金森病(PD)模型小鼠黑质-纹状体神经元的保护作用。方法取PD模型小鼠40只,随机分为淫羊藿组、女贞子组、黄精组、司来吉兰组及模型组5组,每组8只,另取同周龄C57BL/6J雄性小鼠8只作为正常对照组。正常对照组及模型组均给予蒸馏水,淫羊藿组、女贞子组、黄精组、司来吉兰组分别灌胃给予淫羊藿、女贞子、黄精、司来吉兰的水煎液及水溶液。每次灌胃0.5ml,2次/d,连续给药4 w后冰浴中取黑质-纹状体,流式检测FasL、Fas、Caspase-3、Bcl-2,酶联免疫法检测神经生长因子(NGF)、脑源性神经因子(BDNF)、胶质源性GDNF,电镜下观察小鼠黑质中神经元的形态。结果黄精组FasL的含量明显低于模型组(P<0.05),与司来吉兰组及正常对照组无明显差异(P>0.05);淫羊藿组、女贞子组及黄精组的Caspase-3的含量均低于模型组(P<0.05),均与正常对照组和司来吉兰组无明显差异(P>0.05);黄精组中NGF的含量明显高于模型组、司来吉兰组(P<0.05),与正常对照组无明显差异(P>0.05);淫羊藿组和黄精组中的BDNF的含量均高于模型组和正常对照组(P<0.05),淫羊藿组与司来吉兰组无明显差异(P>0.05),黄精组明显高于司来吉兰组(P<0.05);淫羊藿组、女贞子组及黄精组小鼠黑质神经元凋亡程度较模型组有减轻。结论补肾中药可能通过降低黑质-纹状体中相关凋亡因子的含量、增加NGF的含量来对DA能神经元起保护作用的。 Objective To explore the protective effects of Chinese nourishing kidney herbs on neurons of striatal neurons in Parkinson′s disease(PD) model mouse.Methods 40 PD model mice were randomly divided into five groups: epimedium,ligustrum lucidum,polygonatum,selegiline and model control groups.8 normal male C57BL/6J strains of mice were selected into normal control group.The normal control and model control groups were administrated with water,the mice in epimedium group,ligustrum lucidum group,polygonatum group and selegiline group were administrated with epimedium,ligustrum lucidum,polygonatum and selegiline water decoction 0.5 ml,bid,respectively.After 4 weeks,striatal neurons were quickly separated on the ice.Flow cytometry analysis was used to detect the level of caspase-3,Fas,Fas-L and bcl-2.ELISA was used to detect the level of GDNF,NGF,BNGF and observe cell morphology on microscopy electron.Results The level of FasL in polygonatum group was significantly lower than that of model control group(P0.05),not significantly lower than that of selegiline group and blank control group(P0.05);The level of Caspase-3 in epimedium,ligustrum lucidum and polygonatum groups was lower than that of model group(P0.05),not significantly higher than that of blank control and the selegiline groups(P0.05). Compared with selegiline and model groups,the level of NGF in polygonatum group had increased(P0.05),not increased compared with the blank control group(P0.05).The level of BDNF were significantly higher in epimedium and polygonatum groups than that of model and blank control groups(P0.05),while the polygonatum group had higher level than the selegiline group(P0.05);The extent of apoptosis in treatment group had improved compared with model group.Conclusions The protective effects of Chinese nourishing kidney herbs on neurons of striatal neurons is possibly through decreasing apoptotic factors and increasing NTFs.
