榉树遗传变异分析及优良单株选择 被引量:25

摘要 研究选择同一区域内的23个榉树单株,观察其叶色的变化并测定变色期叶片中花青素的含量;同时利用ISSR分子标记技术对23个榉树单株种质资源进行了遗传基础的差异分析,多态性PCR扩增结果表明:在128个位点中,其中多态位点有116个,多态位点百分率为89.9%;POPGENE32软件计算结果表明,这23个榉树材料平均有效等位基因数为1.5930,Nei遗传多样性指数平均为0.3394,平均Shannon信息指数为0.5007;通过聚类分析将这23个榉树品种分为4组,该聚类结果与之前的形态分类结果有一定的相似性,为榉树优良单株的选育提供理论基础。 In this study,twenty three varieties of Zelkova in the same area were measured at different developmental stage,the changing foliage color were detected and the determinations of anthocyanin content in the leaves through various stages were operated. Genetic diversity and similarity were analyzed in molecular level and the purpose of this study is further perfecting the evaluation of species resource in Zelkova schneideriana.The result showed that a total of 128 bands based on 20 primers were detected,out of which 116 were polymorphic(89.9%).As analyzed by POPGENE32,the average value of effective number of alleles,Nei's gene diversity and shannon's information index were 1.5930,0.3394,0.5007,respectively.Clustering analysis showed that the 23 varieties of Zelkova schneideriana could be distinctively classified into four groups.This result can provide theoretical basis for breeding of Zelkova schneideriana single plant.
