摘要
目的确定ZTC1+1-Ⅱ型澄清剂应用于莲子心提取液除杂的最佳工艺。方法以固形物去除率、甲基莲心碱保留率为指标,分别考察药液浓度、澄清剂加入的顺序、澄清剂的用量、加入澄清剂时的温度、搅拌时间及水浴保温时间对除杂效果的影响。在单因素分析的基础上应用正交实验设计优化除杂工艺条件。结果在药液浓度为1∶6,ZTC1+1-Ⅱ型澄清剂2组分加入顺序为先B后A,ZTC1+1-Ⅱ2组分(浓度为1%)用量分别为0.8 mL.g-1(B)及0.4 mL.g-1(A),作用温度为80℃,分别搅拌10 min后保温30 min的工艺条件下,固形物去除率为22.81%、甲基莲心碱含量保留率为89.42%。结论 ZTC1+1-Ⅱ型澄清剂可用于莲子心提取液的除杂工艺,效果良好。
Objective To determine the optimum precipitation process of the extract of plumula nelumubinis by ZTC1+1- II natural clarify agents. Methods Solid removal rate and remaining rate of neferine were used as indexes. The effect of concentration of the extracts, adding order, dosage, reaction temperature and time of clarifier was studied. On the basis of single-factor test, orthogonal design was used to optimize the precipitation process. Results The optimal process was condensing the extract solution to 1 : 6 (herb : herb solution), and the adding order of 2 components of ZTC1+1-II natural clarifier was component B before component A. The dosages of ZTC1+1- II were 0. 8 mL · g-1 for clarifier B and 0.4 mL · g-1 for clarifier A, the reaction temperature was 80℃, and then stirring for 10 min and holding in water bath at 80℃ for 30 min respectively. The solid removal rate was 22.81% and the remaining rate of neferine was 89. 42 %. Conclusion ZTC1 + 1- II natural clarify agents can be used to effectively remove the impurity in plumula nelumubinis extract.
