江淮流域小麦赤霉病菌的遗传多样性 被引量:8

Genetic Diversity of Fusarium spp. Isolates in Yangtze-Huaihe River Valley

摘要 小麦赤霉病是温暖潮湿和半潮湿地区麦类作物的重要病害。小麦感染赤霉病后,不仅减产严重、品质变劣,而且病菌产生的毒素还会对人畜健康产生危害。引起小麦赤霉病的镰刀菌至少有20种,鉴定病原菌及种群结构对赤霉病的防治具有重要意义。针对我国江淮流域江苏、河南、安徽、湖北省的赤霉病菌株,利用7对种属特异性引物进行种属鉴定,在此基础上依据RAPD和AFLP分子标记进行遗传多样性分析。研究结果表明,所有供试菌株均属于禾谷镰刀菌(Fusarium graminearum Schw),分子标记Shannon多样性指数(I)以及Nei氏基因多样度(h)均显示江淮流域禾谷镰刀菌具有较为丰富的遗传多样性。遗传标记相似系数变异范围为0.57~0.99,表明菌株间存在明显的遗传多样性。以相似系数0.63为阈值,可将供试菌株划分为3类,亚类分析表明江淮流域收集菌株具有一定的地域性分布。 Wheat Fusarium head blight (FHB),caused by Fusarium spp.,is one of the worldwide destructive diseases of wheat in the warm,semi-humid and humid regions. The disease not only causes significant yield losses and grain quanlity reduction,but also induces toxin in contaminated seeds,which is harmful to human beings and livestocks. Identification of Fusarium species and understanding genetic population have great significance to prevention and cure of FHB since FHB can be induced by at least 20 species. In the present study,32 single-spore isolates of Fusarium spp. collected and purified from Jiangsu,Henan,Anhui and Hubei Provinces were identified by species-specific PCR detection. Randomly amplified polymorphic DNA (RAPD) and amplified fragment length polymorphic (AFLP) markers were used to evaluate the genetic diversity among Fusarium isolates. The results indicated that all isolates belonged to F. graminearum since they could amplify PCR products only by F. graminearum specific primes pairs Fg11. Isolates expressed high genetic diversity in percentage of polymorphic bands (PPB),Shannon's information index (I) and Nei's gene diversity (h) calculated by software PopGene version 1.32.The marker-based genetic similarity (Gs) among F. graminearum isolates ranged from 0.57 to 0.99 with a mean of 0.82 indicating high degree of genetic diversity in the population. The dendrogram constructed after polymorphic marker data revealed that Fusarium population could be classified in to 3 groups at 0.63 similarity coefficient. Furthermore,formation of subgroups suggested that genetic diversity was related to geographic separation.
