摘要
目的从噬菌体随机12肽库中筛选TGF-β1Ⅱ型受体(TGF-beta receptorⅡ,TβRⅡ)的特异性序列,评估其对瘢痕疙瘩成纤维细胞的作用。方法以人TGF-β1单克隆抗体为靶,在噬菌体随机12肽库中进行4轮生物筛选;应用ELISA法挑选结合活性较好的单克隆噬菌体,进行DNA序列分析;MTT法评估TGF-β1单克隆噬菌体对瘢痕疙瘩成纤维细胞的生物学效应。结果测序共获得5种类似于TβRⅡ的特异性序列。MTT结果显示其中有3种噬菌体模拟肽能够抑制瘢痕疙瘩成纤维细胞增殖。结论具有TβRⅡ特异性序列的噬菌体模拟肽能够抑制瘢痕疙瘩成纤维细胞增殖。
Objective To isolate the specific sequence of TGF-β receptor Ⅱ(TβRⅡ) from a phage 12-mer display peptide library and to evaluate their effects on keloid fibroblasts.Methods A phage 12-mer display peptide library was screened for four rounds using monoclonal anti-human TGF-β1 as the target.ELISA was performed to select monoclonal phages with good association for DNA sequencing.MTT assay was used to detect the effects of monoclonal phages on keloid fibroblasts.Results Five specific sequences were obtained,which were similar to TβRⅡ.The results of MTT assay showed that three of the five phage model peptides could inhibit proliferation of keloid fibroblasts.Conclusion Phage model peptides with specific sequences of TβRⅡ can inhibit proliferation of keloid fibroblasts.
