摘要
血、肝、尿中地西泮、去甲西泮、替马西泮及奥沙西泮用GDX301大孔树脂萃取,萃取物制成酸性水溶液测定二阶导数光谱,方法简便快速.血和尿中4种药物的提取率近似为100%,检出限近似为0.8mg/L;肝中4种药物的提取率近似为80%,检出限近似为1.2μg/g.
The urine, the dilution of blood with water or the supematiant of liver sample being deproteined with 6% HClO4 solution was added GDX301 porous polymer bead and shaken to absorb the drugs in specimen. Then the bead was packed into a glass tuble collumn. The bead was washed with water and elution was effected with dichloromethane. The eluate was evoprated to dry, the residue was dissolved with 0.1 mol/L hydrochloride acid and submitted to second derivative UV spectrophotometry over the range 230 to 280 nm. Calibration graph of drugs was linear over the range 0.5 to 5 mg/L, thedetection limits of drugs in blood, liver and urine were about 0. 8 mg/L, 1.2μg/g and 0. 8 mg/Lrespectivley and the extraction effeciency of drugs in blood, liver and urine were 100% , 80% and 100% respectively. The measurements of this method were the total amount of drug and its metabolites for actual poisoning specimen. For exemple if diazepam had been intaken the measurements of the drug in specimen was the total amount of diazepam and its metabolites, nordiazepam, temazepam and oxazepam.
