摘要
目的:建立HPLC测定血清中地西泮的浓度的方法。方法:采用高效液相色谱法,色谱柱为C18柱(4.6mm×150mm,5μm),柱温:30℃,检测波长:254nm,流动相为甲醇-水(65:35),流速为1mL/min。结果:在0.2—4μg/mL范围内线性良好,R=0.9991,在低、中、高浓度下的回收率为103.9%、104.3%、95.8%,日内、日间RSD均<6%。结论:该方法简便、快速,适合用于临床治疗药物监测。
Objective: To establish the method for the determination of Diazepam in serum by HPLC. Methods: HPLC was used. C18 column (4.6mm × 150mm, 5μm) was adopted, and the temperature was set at 30 ℃, The UV detector was operated at 254nm, the mobile phase consisted of methanol - water (65:35), the flow rate was 1mL / rain. Results: The calibration curve was linear in the concentrations range from 0.2 to 4μg/mL (R=0.9991). The method recovery rates of low, moderate and high concentrations were 103.9%, 104.3% ,95.8%. RSD of intra-day and inter-day was less than 6%. Conclusion: The method is convenient,rapid, suitable for determination of Diazepam in serum.
