三种枸杞组培快繁技术的研究 被引量:3

摘要 以枸杞、新疆枸杞、宁夏枸杞的种子为外植体进行组培快繁技术的研究,结果表明,通过组培法获得的外植体污染率低,而水培法获得的外植体污染率高。枸杞最适宜的初代培养基为MS+6BA 1.5mg.L-1+IBA0.2 mg.L-1,宁夏枸杞和新疆枸杞最适宜的初代培养基为MS+6BA 2.5 mg.L-1+IBA0.2 mg.L-1;三种枸杞最佳的继代培养基均为MS+6BA 0.5 mg.L-1+IBA1.0 mg.L-1,分化诱导率分别为60%、40%、44%,增殖倍数分别为4.5倍、3.0倍、3.0倍;在MS+6BA 0.5 mg.L-1+IBA0.1 mg.L-1培养基上三种枸杞的幼苗生长均表现为粗壮;生根培养基以1/2MS+IBA0.1 mg.L-1效果最佳。以腐殖土1∶蛭石1∶河沙1等量混合配制的移栽基质效果最佳,移栽成活率为96%。 Seeds of Lycium chinense,Lycium barbarum and Lycium barbarum were cultured in tubes.The results were as follows: Contamination rate of explants from tissue culture was lower than those from water culture.the optimum primary culture medium for L.chinense was MS+ 6BA 1.5 mg·L-1+IBA0.2 mg·L-1,and MS+ 6BA 2.5 mg·L-1+IBA0.2 mg·L-1 was for L.barbarum and L.barbarum.The optimum successive transfer culture for all three species was MS+6BA 0.5 mg ·L-1+IBA1.0 mg·L-1,with which the rate of induction and differentiation were 60%,40%,and 44%,and the proliferation rate were 4.5,3.0 and 3.0 times,respectively.The seedlings of three species grew vigorously in medium of MS+6BA 0.5 mg·L-1+IBA0.1 mg·L-1.The optimum rooting medium was 1/2MS+IBA0.1 mg·L-1.The best medium for transplantation found in this research was humus soil,vermiculite and river sand at 1∶1∶1,in which the survival rate of transplantation reached to 96%.
