3种药用枸杞过氧化物同工酶遗传多样性研究 被引量:4

On the Genetic Diversity of Peroxidase Lsozymes of Three Medical Species of Lycium Genus

摘要 利用垂直板式聚丙烯酰胺凝胶电泳技术,检测了中国产枸杞属(Lycium Lirm,)3个药用种(枸杞、宁夏构杞、新疆枸杞)6个居群过氧化物酶,对酶谱进行了分析,计算出基因频率、多态位点比例(P)、各位点等位基因平均数(A)、期望杂合度(He)、固定指数(F)、遗传一致度(Ⅰ)并进行了聚类分析和遗传多样性分析.结果表明:枸杞过氧化物同工酶谱带显示5个位点,每个位点平均等位基因数量为1.4,1.6,1,25;3种枸杞多态住点比例分别为60%,80%,60%;平均杂合度分别为0.5728,0.3249,0,2594,枸杞和宁夏枸杞之间遗传一致度较低(0.5718~0.5988);宁夏枸杞和新疆枸杞、河北枸杷的遗传一致度却比较高(0.6815—0.8197).3种枸杞居群遗传变异类型为HT=0.6555。GST=0.3178,即总的基因变异的31.78%来自于居群间,68.22%来自于居群内. Peroxidase isozymes diversity of 6 populations of three medical species of Lycium genus in China, which were Lycium chinense Mill, L. barbarum L. and L. dasystemum Pojark., was studied by polyacrylamide gel electrophoresis method. The frequency of gene, polymorphic loci(P), average number of alleles per locus(A) and mean expected heterozygosity (He), fixed index(F) and identify of heredity of populations(Ⅰ) were calculated through analysis isozyme belts. Genetic diversity of populations was measured and populations' cluster was analyzed through identify of heredity. The results showed that: there are five loci of peroxidase isozymes of the three medical species of Lycium genus, and mean number of allele(A) of populations were 1.4, 1.6 and 1.25 separately. The percentage of polymorphic loci and mean expected heterozygosityof L. chinense Mill, L. barbarum L. and L. dasystemum Pojark. were 60%,80% and 60% ; 0.572 8, 0.324 9 and 0.259 4"separately. Identify of heredity between L. chinense Mill and L. barbarum L. was lower (0. 571 8 - 0.0. 598 8) and that between L. barbarum L. and L. dasystemum Pojark. was higher(0. 681 5-0. 819 7). The model of genetic diversity of populations of three species was HT = 0. 655 5, GST = 0. 317 8, that was the total genetic diversity came from within populations(68.22% ) and without populations(31.78 % ).
