猪囊虫病基因工程疫苗的研制 被引量:9

摘要 从猪带绦虫六钩蚴cDNA 文库中筛选目的基因并进行克隆表达,重组抗原用血清学方法和猪体免疫试验进行鉴定。将具有免疫保护作用的重组抗原纯化,与免疫刺激复合物疫苗佐剂混合,制备成猪囊虫病基因工程疫苗,免疫仔猪并攻虫。该疫苗安全、无毒副作用,免疫猪减虫率92.2% ,完全保护率55.5% ,且免疫组发现的囊虫多数已死亡。以上结果表明用大肠杆菌表达的重组抗原制备的猪囊虫病基因工程疫苗安全、高效、成本低廉、可规模化生产,有望成为预防猪囊虫病的一种新型生物制剂。 The purpose genes were screened from the cDNA libray of Taenia solium oncospheres, subcloned and expressed.The expressed recombinant antigens were identified by serological method and immunized pigs, respectively. Piglets were immunized with the genetic engineering vaccine made of the purified protective antigen, whichs adjuvant was ISCOM, and challenged orally by viable T. solum eggs, the immunical protective effects were observed by calculating the cysticerci number. The vaccine was safety, no toxic and side effects to piglets, the protective rate against the cysticerci was 92.2% and the protective rate of piglets was 55.5% in constrast to the nonvaccinated group. Most of the cysticerci found in the immunized group were dead. These results revealed that the genetic engineering vaccine against swine cysticercosis, which was made of the recombinamt antigen expressed in E. Coli, was satety, effiency,low price and scaled productivity ,it would be a new biological preparation for preventing swine cysticercosis.
