c-myc的反义寡核苷酸抑制血管平滑肌细胞增殖的研究

摘要 目的探讨c-myc的反义寡核苷酸抑制培养大鼠血管平滑肌细胞增殖的研究。方法 贴壁法培养血 管平滑肌细胞,将不同浓度的反义及正义寡核苷酸加入培养液,观察对血管平滑肌细胞增殖的作用。结果5μg/ml c-myc的反义寡核苷酸加入培养液后第 5、7天抑制血管平滑肌细胞增殖,15μg/ml c-myc的反义寡核苷酸加入培养液 后第3、5、7天抑制血管平滑肌细胞增殖。各浓度c-myc的正义寡核苷酸未发现其抑制血管平滑肌细胞增殖。结论 c-myc的反义寡核苷酸抑制培养大鼠血管平滑肌细胞增殖。 Objective To investigate the effect of antisense c-myc oligodeoxynucleotides on smooth muscle cell (SMC) proliferation in vitro Methods : SMC was isolated from thoracic-abdominal aortic explants of wistar rats The cells were cultured in DMEM containing 10% FBS. Oligodeoxynucleotides were added to culture medium and the effect on SMC proliferation was observed ,Results 5μg/ml antisense c-myc oligodeoxynucleotides inhibit SMC proliferation after adding it to the medium at the fifth and seventh days. 15μg/ml antisense c-myc Oligodeoxynucleotides inhibit SMC proliferation after adding it to the mediuxn at the third, fifth and seventh days. We didn't observe the effect of sense c-myc Oligodeoxynucleotides on SMC proliferation Conclusions Antisense c-myc oligodeoxynucleotides inhibit; SMC Proliferation in vitro.
