碱性成纤维细胞生长因子和白细胞介素6促进大鼠血管平滑肌细胞增殖的协同作用 被引量:8

The Synergistic Effect of Basic Fibroblast Growth Factor and Interleukin-6 on Rat Vascular Smooth Muscle Cell Proliferation

摘要 为观察碱性成纤维细胞生长因子和白细胞介素 6促进血管平滑肌细胞增殖的协同作用 ,采用大鼠主动脉贴块法培养血管平滑肌细胞 ,用 5 -溴 - 2 -脱氧尿嘧啶掺入法和细胞计数法观察碱性成纤维细胞生长因子和白细胞介素 6对平滑肌细胞增殖的影响。结果发现 ,0~ 10 .0 μg/L碱性成纤维细胞生长因子和 0~ 10 0 μg/L白细胞介素 6能促进平滑肌细胞增殖 ,且呈剂量依赖性 ,1.0 μg/L碱性成纤维细胞生长因子和 1.0 μg/L白细胞介素 6促进细胞增殖 ,且 48h内呈时效依赖性 ,作用高峰在 2 4h ,两者合用促进细胞增殖作用大于两者单用作用之和 ,呈协同作用。 Aim To observe the synergistic effect of basic fibroblast growth factor and interleukin-6 on vascular smooth muscle cells proliferation. Methods BrdU incorporation and cell counting method were adopted to observe the effects of basic fibroblast growth factor and interleukin-6 on the proliferation of rat aortic cultured smooth muscle cells. Results 0~10.0 μg/L basic fibroblast growth factor and 0~100 μg/L interleukin-6 were able to promote smooth muscle cells proliferation separately, and synergistically in combination in dose- and time-dependent manner. The pro-proliferation effect reached climax at 24 hours. Conclusion Basic fibroblast growth factor and interleukin-6 had synergistic effect on vascular smooth muscle cells proliferation.
