摘要
目的：观察低剂量5-氟脲嘧啶（5—FU）持续输注联合低剂量顺铂（PDD）方案治疗晚期消化道恶性肿瘤的疗效。方法：5－FU250mg．m－2．d－1，持续深静脉输注3～4周。PDD6mg．m-2．d-1，静脉输注1小时，每周用5天，连用3～4周。以上方案连用二周期为一疗程，化疗后休息1个月评定疗效。结果：全组24例，总有效率为54．17％，其中PR13例。胃癌、食管癌、结肠癌的有效率分别为：64.29％、50．00％、33．33％；肝转移灶的有效率较高为75．00％。中位缓解期4．5个月（2～8个月）。没有Ⅲ°～Ⅳ°毒副反应发生。结论：认为低剂量5－FU持续输注联合低剂量PDD方案治疗晚期消化道恶肿瘤是一个有效低毒的化疗方案。
Objectives:To evaluate the efficacy and toxicity of the continuous central venous infusion(c. c. v. i) of low-dose of 5-fluorouracil with low-dose of cisplatin (PDD). Methods: Twenty-fourpatients with advanced or recurrent gastrointestinal carcinoma (14 in stomach, 4 in esophagus,and 6 incolon)were observed in this study. 5-Fu was given with a dosage of 250mg· m-2·' d-1. c. c. v. i. for 34 weeks,and PDD with a dosage of 6mg· m-2· d-1. infused in one hour,five times a week, and lasted for3-4weeks. Two treating courses were carried out with a pne month interval in between. Results: Theoverall response rate was 54. 17% (13/24), including 13 cases of partial responses. The response ratesof gastric, esophageal, and colonic cancers were 64. 29% (9/14), 50. 00% (2/4) and 33. 33% (2/6 )respectively. An especially high response rate of 75% was seen in 12 cases with liver metastasis. Themean duration of response was 4. 5 months (range 2-8 months). No severe side effects occurred in anyof the cases. Conclusions: It is indicated that a combined chemotherapy of daily low- dose of 5-Fuwith PDD is an effective method in the treatment of advanced or recurrent gastrointestinal carcinoma.
