摘要
目的:研究牙龈卟啉单胞菌脂多糖(Porphyromonas gingivalis lipopolysaccharide,P.gingivalisLPS)在氧化低密度脂蛋白(oxidized low density lipoprotein,oxLDL)刺激巨噬细胞形成泡沫细胞过程中和形成泡沫细胞后对NF-κB信号通路的影响。方法:用oxLDL诱导THP-1人单核细胞源性巨噬细胞形成泡沫细胞,在巨噬细胞和泡沫细胞中加入P.gingivalisLPS和oxLDL,利用免疫细胞化学技术检测NF-κB p65亚单位核转位情况,Western blot检测细胞质内IκB-α变化。结果:P.gingivalisLPS能促进巨噬细胞和泡沫细胞中p65亚单位核转位,在2h时达到最高水平;也能降低细胞质内IκB-α水平,在2h时达到最低水平;在2h时,10μg/ml比10、100ng/ml、1μg/mlP.gingivalisLPS更能提高p65核转位率。结论:P.gingivalisLPS可以促进泡沫细胞形成过程中和形成后NF-κB信号通路的激活,促进动脉粥样硬化进展。
Objective:To investigate the effect of Porphyromonas gingivalis lipopolysaccharide(P.gingivalis LPS)on NF-κB pathway in oxidized low density lipoprotein(oxLDL)incubated macrophages during and after foam cells formation.Methods:Macrophages from THP-1 human monocytes were incubated with oxLDL to generate foam cells.P.gingivalis LPS were added with oxLDL into the culture of macrophages and foam cells.NF-κB p65 subunits nuclear translocation was evaluated with immunocytochemistry,IκB-α in cytosol was detected by western blot.Results:P.gingivalis LPS enhanced NF-κB p65 nuclear translocation in macrophages and foam cells with the highest translocation rate at 2 h,and decreased IκB-α level in the cytoplasm with the lowest level at 2 h.10 μg/ml P.gingivalis LPS demonstrated highest p65 translocation rate when compared with 10 ng/ml,100 ng/ml,1 μg/ml LPS.Conclusion:P.gingivalis LPS can enhance NF-κB pathway activation during and after foam cells formation.
