口腔黏膜白斑及鳞癌组织中核因子-κB p65与PCNA的表达 被引量:2

摘要 目的:探讨口腔黏膜白斑(OLK)及鳞癌(OSCC)组织中核因子-κBp65及增殖细胞核抗原(PCNA)的表达情况。方法:应用免疫组织化学技术检测正常口腔黏膜上皮(n=10)、单纯增生型OLK(n=11)、异常增生型OLK(n=35)、OSCC(n=36)组织中的NF-κB p65和PCNA的表达情况。结果:正常口腔黏膜上皮组织中p65不表达,由单纯增生型OLK转变到异常增生型OLK最后到OSCC的发生发展过程中NF-κB p65的表达明显上调(9.1%,42.9%,72.2%),差异有统计学意义(χ2=30.323,P<0.05);正常口腔黏膜、OLK组织和OSCC组织中均有PCNA阳性表达,且增殖指数逐渐升高(4.90±1.18,6.45±2.03,28.43±13.91,63.89±15.31),差异有统计学意义(F=21.55,P<0.05)。二者在OLK癌变过程中密切相关。结论:二者可能是检测OLK恶变潜能及OSCC早期诊断的敏感指标。 Aim : To investigate the expression of NF-KB p65 and PCNA as well as their relation in Oral Leukoplakia (OLK) and oral squamous cell carcinoma (OSCC) ,then to evaluate their functions in tumorigenesis and progession of OS- CC. Methods:Immunohistochemical staining technique was used to determine the expression NF-KB p65 and PCNA in all specimens: 10 cases of normal human oral epithelium, 1 lsimple hyperplasis OLK,35 dysplasia OLK, and 36 OSCC. Resuits: The positive rates of NF-KB p65 in normol oral mucosa, simple hyperplasia OLK, dysplasia OLK and OSCC were 0% , 9.1% , 42.9% , and 72.2% respectively, and a statistical significance was analysed( P 〈 0.05 ). PCNA index in normol oral mucosa, simple dysplasia, dysplasia and O5CC were 4.90 ± 1.18 , 6.45 ± 2.03, 28.43 ± 13.91 and 63.89 ± 15.31. There were significant differences between expression of p65 and PCNA-LI in OLK specimens ( t = 3. 121, P 〈0.05) and in OSCC specimens (t =2. ll6,P 〈0.05). Conclusion: NF-KB p65 and PCNA protein may be useful marks for evaluating the malignant potential of OLK and early diagnosis of OSCC.
