摘要
用高效液相色谱法对氟哌啶醇、氯氮平、阿普唑仑、丙咪嗪、阿米替林、氯丙嗪、泰尔登、氟奋乃静、地西泮等9种常用精神病治疗药物过量中毒进行定量分析。标本经一步液-液提取后,在C8柱上进行分离,流动相为甲醇-乙腈-水,紫外检测波长为254nm,9种药物在17min内即可实现良好分离,线性范围为0.5~16μg/ml,绝对回收率为76.8%~93.3%,最低检测浓度为0.1~0.4μg/ml,日内和日间精密度小于7.5%。本法具有简便灵敏、快速准确的优点,适合于临床检测。
HPLC method for determination of nine psychotherapeutic drugs.haloperidol,clozapine,alprazolam,amitriptyline,imipramine,chlorpromazine,diazepam,chlorprothixene,fluphenazine in plasma was developed.The sample was extracted by one step liquid liquid extraction followed by separation on C 8 column.The mobile phase was a mixture of methanol∶acetonitrile∶water ( 16.0 ∶ 36.5 ∶ 47.5 ).The UV detector wavelength was set at 254nm.Nine drugs were effectively separated in 17 minutes.The standard curves of all nine drugs were linear over the range of 0.5  ̄16μg/ml.The absolute recoveries were ranged from 76.8% to 93.3% .The detection limits were in the range of 0.1  ̄ 0.4 μg/ml.The within day and between day RSD were less than 7.5% .This method is simple,sensitive,rapid and accurate.It is satisfactory for the determination of drug overdose.
