摘要
目的探讨淋巴细胞功能相关抗原1(LFA-1)的单克隆抗体对大鼠肝脏移植后转化生长因子-β1(TGF-β1)表达的影响。方法将肝脏移植后的大鼠随机分为第1组(他克莫司组),第2组(LFA-1单抗组)和第3组(他克莫司+LFA-1单抗组)。于术后100d获取移植物作病理学检查,对慢性排斥反应进行评分,并检测移植物中TGF-β1的表达。结果第3组的慢性排斥反应明显轻于第1组(P=0.004),而第3组移植物中TGF-β1的阳性表达明显低于第1组(P<0.05)。结论 LFA-1的单克隆抗体可抑制大鼠肝脏移植后TGF-β1的表达。
This paper aimed to evaluate the role of lymphocyte function-associate dantigen1（LFA-1）monoclonal antibody on expression of TGF-β1（transforming growth factorβ1） after liver transplantation in rats.Rats were underwent liver transplantations,an then randomly divided into tacrolimus group,LFA-1 monoclonal antibody group,and tacrolimus＋LFA-1 monoclonal antibody group.One hundred days after transplantations,the grafts were harvested for histological examination,and the score of chronic rejection was evaluated.At the same time,the expression of TGF-β1 in grafts was measured.The score of chronic rejection in tacrolimus ＋ LFA-1 monoclonal antibody group was lowerthan that of tacrolimus group（P = 0.004）;the expression of TGF-β1 in grafts of tacrolimus ＋ LFA-1 monoclonal antibody group was lowerthan that in tacrolimus group（P 0.05）.We conclude that LFA-1 monoclonal antibody may inhibit the expression of TGF-β1 in graft afterlivertransplantation in rats.
