环磷腺苷葡胺预处理对大鼠脑缺血再灌注损伤后细胞凋亡的影响 被引量:1

摘要 目的研究环磷腺苷葡胺(MAC)预处理对大鼠局灶性脑缺血再灌注(CIR)损伤后细胞凋亡的影响。方法 50只SD大鼠随机分为正常对照组(5只)、假手术组(5只)、CIR组(20只)和MAC预处理组(20只),CIR组与MAC预处理组又分为缺血2h后再灌注2h、24h、48h和7d4个亚组,每个亚组5只大鼠。MAC预处理组制模前3d给予MAC5mg/(kg·d)腹腔注射。线栓法建立大鼠CIR模型,流式细胞术检测缺血侧海马神经细胞凋亡。结果与正常对照组比较,CIR组与MAC预处理组在CIR24h~7d时细胞凋亡率均显著升高(均P<0.01)。与CIR组比较,CIR24h、48h时MAC预处理组细胞凋亡率明显降低(均P<0.01)。与CIR7d时比较,CIR2h时细胞凋亡率明显降低,而CIR24h、48h时明显升高(均P<0.01)。结论 CIR大鼠细胞凋亡显著增加,MAC预处理能有效抑制海马神经细胞凋亡,对CIR大鼠有保护作用。 Objective To study the effects of meglumine adenosine cyclosphosp (MAC) preconditioning on apoptosis after cerebral ischemic reperfusion (CIR) in rats. Methods Fifty SD rats were randomly divided into normal control group (n = 5) ,sham operation group (n = 5) ,CIR group (n = 20) and MAC preconditioning group (MAC group,n = 20) . CIR group and MAC group were separately divided into 4 sub-groups with CIR 2 h,24 h,48 h and 7 d after ischemia 2 h,and each sub-group had 5 rats. Rats in MAC group were given 5 mg /(kg·d) MAC by intraperitoneal injection at 3 d before operation. The rat models of CIR were established by suture embolism method. The apoptosis of neuron of hippocampus was detected by flow cytometry. Results Compared with normal control group,the apoptosis rates of CIR group and MAC group were significantly increased at 24 h - 7 d after CIR (all P 0. 01) . Compared with CIR group,the apoptosis rate of CIR group was significantly decreased at 24 h,48 h after CIR (all P 0. 01) . Compared with CIR 7 d sub-group,the apoptosis rate was significantly decreased at 2 h after CIR,while significantly increased at 24 h,48 h after CIR (all P 0. 01) . Conclusions The apoptosis of CIR rats increased significantly. MAC preconditioning can inhibit the apoptosis of neuron of hippocampus,and have protective effects on CIR rats.
