摘要
开发一种分析2-去氧-D-葡萄糖样品的离子色谱方法,该方法可准确定量2-去氧-D-葡萄糖样品的含量.经色谱分离条件优化,选用DIONEX CarboPac PA10(4 mm×250 mm)色谱柱,以18 mmol/L NaOH溶液为流动相,流速为0.8 mL/min,用脉冲安培检测器检测,实现2-去氧-D-葡萄糖、葡萄糖和氨基葡萄糖3种结构极为相似单糖的高效分离,建立起2-去氧-D-葡萄糖的离子色谱法.2-去氧-D-葡萄糖在0.618~61.8μg/mL范围内呈现良好的线性关系,相关系数为0.998 6,最低检测限为27.2μg/L,最低定量限为216.3μg/L,平均回收率101.3%,实验结果表明该离子色谱法检测2-去氧-D-葡萄糖,简单、灵敏,测定结果准确,是测定2-去氧-D-葡萄糖含量的有效方法.
An effective ion chromatographic method was developed to quantitative analyse 2-deoxy-D-glucose(2-DG).2-DG,glucose and amino-glucose are three monosaccharides with remarkably similar structures.So it is difficult to separate 2-DG and to detect the content of 2-DG accurately.By optimizing chromatographic separation conditions,the high efficient separation and quantitative analyse method of 2-DG was established.The optimal separation and quantitative analysis of 2-DG was performed on a DIONEX CarboPac PA10 Column(4 mm×250 mm) with a mobile phase of sodium hydroxide solution(c=18 mmol/L).The flow rate was 0.8 mL/min and the detector was pulse ampere detector(PAD).A good linearity was observed in the concentration range of 0.618 μg/mL to 61.8 μg/mL,and the correlation coefficient was 0.998 6.The lowest detection and quantitative limits were 27.2 μg/L and 216.3 μg/L,respectively.The average recovery was 101.3%.These results indicated that ion chromatographic method was a simple,sensitive,accurate and effective method for detection of 2-DG.
