Establishment of Caco-2 Cell Model and Exploration of Its Application to Absorption of Chinese Medicine

摘要 目的建立Caco-2细胞模型并探讨其在中药吸收研究中的应用。方法通过显微镜观察细胞形态学特点、测定跨细胞膜电阻和荧光黄通透量等指标,对Caco-2细胞模型进行评价。结果各项指标的测定值符合要求,本实验建立的Caco-2细胞模型的完整性、紧密性和通透性良好。结论建立的Caco-2细胞模型可用于研究中药化学成分转运的吸收机制;高灵敏度分析仪器可以促进Caco-2细胞模型在中药化学成分吸收研究中最大限度的应用;通过两个或多个中药化学成分相互作用的研究,可以阐明中药配伍和中药复方应用的科学性。 Objective To establish the Caco-2 cell model and to explore its application to the research of absorption characteristics of Chinese Medicine.Methods The Caco-2 cell model was evaluated by morphological feature,transepithelial electrical resistance(TEER) and the transport of lucifer yellow.Results The Caco-2 cell model has been established in this experiment.The determined values met the requirements,and the integrality,permeability and the transportation of lucifer yellow in the cell model were good.Conclusion The established Caco-2 cell model can be used to study the absorption mechanism of chemical components of oral Chinese medicine.High-sensitivity analytical instruments can promote the maximum application of Caco-2 cell model in the absorption research of chemical components of Chinese medicine.The absorption research of interaction between two or more chemical components of Chinese Medicine will supply scientific evidence for compatibility of traditional Chinese drugs and application of compound prescriptions of Chinese Medicine.
