芍药苷油水分配系数的测定及pH对其的影响 被引量:13

摘要 目的:通过处方前研究,考察芍药苷的溶解特性,测定芍药苷在不同pH值缓冲液中的油水分配系数,为芍药苷脂质体的处方设计提供参考。方法:用振摇法测定芍药苷的溶解度及正辛醇-水分配系数,比较pH值对油水分配系数的影响。结果:芍药苷在水、氯仿中的溶解度分别为50.57mgmL-1、68.34mgmL-1;在水、pH值分别为2.0、2.5、4.0、5.0、5.8、6.8、7.8的磷酸盐缓冲液中的正辛醇-水分配系数分别为-0.47、-0.61、-0.76、-0.71、-0.50、-0.78、-0.98、-0.58。结论:芍药苷易溶于水和氯仿;pH对芍药苷的油水分配系数有一定的影响,在pH5.0缓冲盐溶液中的溶解度及油水分配系数最大。 Objective:To provide reference for the prescription of paeoniflorin liposome by investigating the solubility and the n-octanol/water partition coefficients of paeoniflorin and the effect of pH on it. Method:The solubility and n-octanol/water partition coefficients of paeoniflorin were measured by shake-flask method, and the effect of pH on n-octanol/water partition coefficients was investigated. Result:The solubility of paeoniflorin in water and chloroform were 50.57 mg﹒mL-1 and 68.34 mg﹒mL-1, respectively. The n-octanol/water partition coefficients were -0.47, -0.61, -0.76, -0.71, -0.50, -0.78, -0.98, -0.58 in water and phosphate buffers of pH 2.0, 2.5, 4.0, 5.0, 5.8, 6.8, 7.8, respectively. Conclusion:Paeoniflorin is easily soluble in water and chloroform. The n-octanol/water partition coefficients of paeoniflorin are influenced by pH. And the maximum values of solubility and n-octanol/water partition coefficient are found in phosphate buffer of pH5.0.
