摘要
目的:研究山芝麻提取物对肝纤维化大鼠肝组织Ⅰ型胶原(ColⅠ)mRNA和ColⅠ蛋白表达的影响。方法:采用CCl4灌胃复制大鼠肝纤维化模型,6周末用荧光定量PCR法测定山芝麻提取物对大鼠肝组织ColⅠmRNA的相对表达值,用免疫组化法检测ColⅠ蛋白的表达量。结果:山芝麻提取物可显著降低肝纤维化大鼠肝组织ColⅠmRNA和ColⅠ蛋白的表达(P<0.01或P<0.05),且随着药物浓度的增加,其作用逐渐增强,呈现量效反应关系。结论:山芝麻提取物能显著改善肝纤维化大鼠的肝脏组织结构,减轻肝纤维化。其抗肝纤维化的机制可能与抑制ColⅠ有关。
OBJECTIVE：To study the effects of the extract from Helicteres angustifolia on ColⅠmRNA and ColⅠprotein expressions in hepatic tissue of rats with hepatic fibrosis.METHODS：The rat model of hepatic fibrosis was established by intragastric administration with carbon tetrachloride（CCl 4）.At the end of the 6 th week,the expression of ColⅠmRNA in hepatic tissue was determined by Fluorescence quantitative PCR（FQ-PCR）,and the expression of ColⅠprotein was detected using immunohistochemical stain method.RESULTS：Extract of H.angustifolia could markedly decrease the expressions of ColⅠmRNA and ColⅠprotein（P0.05 or P0.01）.As the drug concentration increased,the effects also gradually increased in dose-effect manner.CONCLUSION：Extract of H.angustifolia significantly ameliorates CCl 4 -induced liver fibrosis which is related to the inhibition of the expression of ColⅠ.
