摘要
目的:探索肝细胞在肝纤维化形成中的可能作用。方法:用核酸原位分子杂交方法和3H脯氨酸掺入法,观察原代培养大鼠肝细胞在血小板衍生生长因子(PDGF)刺激前后胶原合成和Ⅰ、Ⅲ型前胶原mRNA的表达及钙拮抗剂维拉帕米对此作用的影响。结果:肝细胞培养一定时间后能够合成胶原,能够表达Ⅰ、Ⅲ型前胶原基因。PDGF能促进肝细胞的胶原合成和前胶原基因表达。但此刺激作用为维拉帕米所抑制。结论:这些研究为肝细胞参与肝纤维化形成提供了实验依据。
Purpose To evaluate the effect of hepatocyte(HC) on liver fibrosis. Methods Effects of platelet derived growth factor (PDGF) and calcium antagonist, verapamil (Ver), on collagen biosynthesis, expression of procollagen types Ⅰ and Ⅲ mRNA of HC were investigated by in situ hybridization and 3H Proline incorporation. Results After HCs were cultured five to seven days, they could synthesize collagen and express procollagen types Ⅰ、Ⅲ mRNA. PDGF could significantly enhance collagen synthesis and the expression of procollagen type Ⅰ、Ⅲ mRNA of HCs and Ver could antagonize the effect of PDGF. Conclusion These findings provide a direct evidence for the participation of HCs in liver fibrosis.
